Subject isolation can be tricky, even with a wide aperture lens, if the background is close to the subject. Two tips for helping to isolate your subject in that situation: First, try to get as close as possible to the subject, this will help blur the background. Secondly, try to find a background that is duller than the subject. There are ways to simulate these effects in post processing, but it is a lot easier to do it while making the image.
